I'm trying to upgrade my JV16 to the latest version and I'm getting the following error message:

C:\Program Files\jv16 powertools\jv16PT.exe

An error occurred while trying to replace the existing file:

DeleteFile failed; code 5.

access is denied

Click Retry to try again, Igonor to skip this file (not recommended)

What should I do?

Maybe the program is still active when you try to update.

Look in Windows Task Manager (Applications and/or Processes) if jv16PT.exe is present and if it is KILL it.

Reboot and try to update again.

The error message means the program file cannot be updated, most likely the program is still running on the background. As Redseujac said, you can try to use Task Manager to kill the process.
