Yes, for example, like this:

Z:\ is the drive in my laptop, mounted on a wireless network.
You can save named setting preferences (per tool) so you don't have to type it all each time. Look at the "Preference saving" tab in the Settings tool.
BTW, multiple full partition searches are usually not advisable (except with Directory Finder) -- File Finder and Duplicate File Finder can take a long, long time to do that.
Also, before you remove anything with Duplicate File Finder, read the manual carefully. It is generally not safe to remove a file just because it is a duplicate.
